Maintenance of Bioburden Testing Environment:

(1) During the test, the windows and air conditioner should be closed, or the room should be disinfected with alcohol spray to avoid the impact of indoor air circulation on the environment in the ultra-clean platform (it is better to operate in a constant temperature and humidity sterile room with a ventilation system ).

(2) If the inspection is carried out in the raw material microbiology inspection room, the ultraviolet light should be turned on regularly to disinfect the room.

Small environment for bioburden detection (clean bench):

1. Regularly clean the primary filter and replace it in time.

2. Before the test, clean the inside of the ultra-clean bench, wipe it with 75% alcohol, and turn on the ultraviolet light of the ultra-clean bench half an hour in advance to disinfect the internal environment of the ultra-clean bench.

3. Turn off the UV lamp, turn on the fan, and adjust the appropriate air intake. The air volume should not be too low.

4. After each test, the inside of the ultra-clean platform must be cleaned and disinfected with 75% alcohol. 5. The UV lamp should be replaced in time according to its service life.

6. At the same time as the detection, an environmental blank for the corresponding colony detection should be made.

7. Selection of detection area:

Generally, aseptic operation is performed in the area 1/3-2/3 away from the outer edge of the ultra-clean platform;

Operate in the outer flame protection area of the alcohol lamp flame.

Cleanliness of Bioburden Testing Instruments:

1. Glassware: Sterilized by dry heat sterilization.

2. Scissors, spoons and other items. Instruments used for testing should be used exclusively for testing and should not be mixed with other operating instruments. When in use, first sterilize with 75% alcohol, and then sterilize with fire.

3. The inoculation needle (ring) is sterilized by burning, but it should be noted that the inoculation needle (ring) should be cooled before testing.

Collection and testing of samples for bioburden testing:

1. Ensure the sterility of the sampling instrument.

(1) Glassware: Use dry heat to sterilize to ensure a constant temperature and sufficient time (but not too long, which may easily cause glassware to break and be scrapped).

(2) Sampling basket: spray with 75% alcohol before use.

(3) Sampling spoon: To ensure its cleaning and disinfection, wipe it with 75% alcohol after cleaning.

(4) Sampling bag: firstly, it can be wiped and disinfected with alcohol, and then sterilized by ultraviolet light in the ultra-clean bench (the packaging workshop should be sterilized by ultraviolet light in the transmission window).

(5) The surface of the electronic scale is disinfected with 75% alcohol.
