1. Packaging and transport parts testing: bioburden testing aseptic operation requirements
1. Wear work clothes and a work cap when inoculating bacteria.
2. When inoculating food samples, you must wear special work clothes, hats and slippers, place them in the buffer room of the sterile room, and use them after ultraviolet disinfection before work.
3. Packaging and transportation test: When inoculating food samples, wash your hands with soap before entering the sterile room, and then wipe your hands with 75% alcohol cotton balls.
Packaging and Shipping Test
4. The straws, plates and medium used for inoculation must be sterilized. Unpackaged utensils should not be stored prior to use. Metal utensils should be autoclaved or burned 3 times with 95% alcohol before use.
5. Packaging and transportation test: When taking the straw out of the package, the tip of the straw should not touch the bare part. When using a pipette to inoculate a test tube or dish, the tip of the pipette should not touch the edge of the test tube or dish.
6. Inoculation samples and bacterial transfer must be carried out in front of an alcohol lamp. When inoculating bacteria or samples, the pipette must be flame sterilized after removal from the packaging and the stopper of the tube has been removed.
7. Before inoculating bacteria, the inoculation loop and inoculation needle should be burned on all wires with a flame, and if necessary, burn to the connection between the inoculation loop, needle and rod. Burned by flames.
8. Packaging and transportation test: When using a pipette to suck up the bacterial liquid or sample, use the corresponding rubber tip to pick up, do not suck it up directly with your mouth.
2. Testing of packaging and transportation parts: requirements for the use of bioburden testing in sterile rooms
1. The windows leading to the sterile room should be sealed with double-layer glass, and should not be opened at will. There should be buffer rooms and sliding doors corresponding to the size of the sterile room, and a small room of 0.5-0.7 square meters. Window for transferring items after entering the sterile room.
2. Packaging and transportation test: The sterile room should be kept clean. After work, use 2%-3% cresol soap to disinfect, wipe the work surface, and do not store items that are not related to the experiment.
3. The door of the sterile room should be closed tightly before and after use, and the ultraviolet light should be turned on. If an indoor hanging ultraviolet lamp is used for disinfection, a 30W ultraviolet lamp is required, the distance is 1.0m, and the irradiation time is not less than 30min. Operate directly under UV rays to avoid damage. The lamp needs to be wiped gently with an alcohol cotton ball every two weeks to remove the dust and grease on it to reduce the impact of UV penetration.
4. Packaging and transportation test: When processing and inoculating food samples, enter the sterile room for operation, and do not enter or exit at will. If you need to deliver items, you can deliver them through a small window.
5. If the air conditioner needs to be installed in a sterile room, there should be a filter device.
3. Testing of packaged and transported parts: bioburden detection, disinfection and sterilization requirements
Bioburden testing Glassware, metalware, culture media, contaminated and inoculated cultures, etc. used for microbial testing must be sterilized before use.
Packaged Shipment Testing: Bioburden Testing Dry Heat and Moist Heat Autoclave Methods
1. Preparation before sterilization
(1) All items that need to be disinfected should be washed and dried first. Glassware such as straws and plates should be tightly wrapped in paper. If metal cylinders are used, the upper ventilation holes should be opened.
(2) Wrap the stopper of the triangular bottle containing the medium with paper, cover the test tube cap, pull out the core from the syringe, and wrap it with gauze.
2. Load
(1) Dry heat sterilizer: The items should not be too crowded and should not touch the walls of the box.
(2) Large-scale high-pressure steam cooker: place and wrap the sterilized items separately, and put them directly into the sterilization tank. The items should not be too crowded.
3. Equipment inspection
(1) Check whether the door switch is flexible, whether the apron is damaged, and whether it is flat.
(2)检查蒸汽排出时压力表是否保持在零位,关上门盖,通蒸汽或热,观察压力锅或立式压力蒸汽灭菌器的使用情况。
